In our facility we have one point of sale cashier site, so we have about 6 departments requiring sales through that one site and each department has different needs. One of them, our Personal Training department, needs us to assign a salesperson to the item being sold so the trainer can get a commission. Our Point of Sale operators don't always remember to assign a salesperson and it can get time-consuming tracking everyone down.

Is there a way to require a salesperson to be defined only for certain items?

Don't you sell personal training as a series sale item?  Do you pay out commissions when the cash is received or when the training is complete?  If you pay out when cash is received, I don't think there is a way to track other than to create a different code for each trainer, which is cumbersome.  Also, I would think that you would have to worry about the trainer reimbursing you if the customer never used the training, or needed a refund, etc, right?  If you pay out after the training is received, if it is a series sale, the trainer can click off of completed sessions and you can track that way, as well as report on unused sessions, expiring sessions, etc.

Not sure if that helps or if I totally missed the point.

To answer your question, there is not a way to require a salesperson at Point of Sale.  You can default the salesperson to being whoever is signed in as the operator, but that doesn't sound like it will help you in this instance.  Perhaps if you gave me a few more details about how your Personal Training commissions work, I can better assist you with this.  Do you use Series Sales for your PT, as Mike was asking?  If so, then you can select a trainer when marking each session as used, and pay them for performing each session.  Or, do you pay commission on the sale of the package in addition to paying for performing each session?
